The cloning process was confirmed by application a PCR using inner primers (TR9 and TR8) after a DNA extraction step based on simple boiling method. Simultaneously, a few transformed colonies were cultured in Luria-Bertani medium containing ampicillin. The plasmid DNA was then extracted by use of GF-1 Plasmid DNA Extraction Kit (Vivantis, Malaysia). by a group of researchers from Ahwaz, Iran.
