Proteinase K is a subtlisin-related serine protease originally isolated from the mold, Tritirachium album. Vivantis new Proteinase K is a recombinant enzyme. The key specifications of the recombinant enzyme are the same as those of the native protease with the amino acid sequence and the molecular structure are identical. Proteinase K hydrolyzes a variety of peptide bonds in a wide range of temperatures and buffers with optimal activity between 20 - 60°C and a pH between 7.5 and 12.0. Proteinase K is also active in buffers containing chelating agents such as EDTA.
The recombinant preparation is much purer than the native enzyme. In particular, since Proteinase K, is DNA-free, it is especially suitable for isolating PCR and RT-PCR templates.
Concentration
20-22mg/ml in 10mM Tris-HCl, pH7.5. The solution contains calcium acetate as stabilizer.
Storage and stability
The undiluted solution is stable at 2 - 8°C. Shipment condition can be room temperature.
Volume activity
50U/mL. One unit is the enzyme activity which cleaves at 25°C in 1 minute 18mmol Chromozym TRY (equivalent to 600U/ml with the haemoglobin assay.)
Specific activity
Approximately 2.5U/mg when assayed with the chromozym assay; equivalent to 30U/mg with the haemoglobin assay
Cleavage specificity
Proteinase K is one of the most active endopeptidases known and does not show any pronounced cleavage specificity. If excess proteinase K is incubated with proteins for a long time, the enzyme will degrade the proteins to free amino acids.
